KCC2 (K, Cl Cotransporter 2)

KCC2 is a member of the cation-chloride cotransporter gene family. It acts as a K-Cl cotransporters. KCCs normally lower intracellular chloride concentrations below the electrochemical equilibrium potential and depending on the chemical concentration gradients of potassium and chloride, KCC2 can operate as a net efflux or influx pathway. It is proposed to act as the main chloride extruder to promote fast hyperpolarizing postsynaptic inhibition in the brain. KCC2 is expressed at high levels in neurons throughout the nervous system and immunofluorescence shows that the protein is localized at inhibitory synapses of the spinal cord. Studies in mice have shown that KCC2 reduces GABA's inhibitory signaling, resulting in motor defects, epilepsy, and anxiety-like behavior.\n\nApplications: \nSuitable for use in Western Blot and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1ug/ml detected KCC2 in rat brain membrane preparations. Rat brain membrane preparation were resolved by electrophoresis, transferred to nitrocellulose and probed with K0120-07. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. \nImmunocytochemistry: 1:200 detected KCC2 in MDCK cells as reported by an independent laboratory.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies: \nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb \nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb I1904-46Q: IgG, H&L (HRP) Pab Gt xRb\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
