The United States Biological Lupine ELISA represents a highly sensitive detection system for lupine and is particularly capable of the quantification of residues in sausage, bakery products, potato products, ketchup and juices. Due to high cross-reactivity the test is suitable for the detection of all commonly used lupine species.\n\nIntended Use:\nFor the quantitative determination of Lupine in food.\n\nSensitivity: 0.2ppm\nRecovery (spiked samples): ~98%-113%\nIncubation Time: 60 min\n\nTest Principle:\nThe United States Biological Lupine quantitative test is based on the principle of the enzyme linked immunosorbent assay. An antibody directed against lupine proteins is bound on the surface of a microtiter plate. Lupine containing samples or standards are added to the wells of the microtiter plate. After 20 minutes incubation at room temperature, the wells are washed with diluted washing solution to remove unbound material. A peroxidase conjugated second antibody directed against lupine proteins is added to the wells and after 20 minutes of incubation the plate is washed again. A substrate solution is added and incubated for 20 minutes, resulting in the development of a blue color. The color development is inhibited by the addition of a stop solution, and the color turns yellow. The yellow color is measured photometrically at 450nm. The concentration of lupine is directly proportional to the color intensity of the test sample.\n\nKit Components:\nL6000-01A: Microtiter plate ,1x96 wells \nL6000-01B: Standard (0ppm/ml), 1x1ml\nL6000-01C: Standard (2ppm/ml), 1x1ml\nL6000-01D: Standard (5ppm/ml), 1x1ml\nL6000-01E: Standard (15ppm/ml), 1x1ml \nL6000-01F: Standard (30ppm/ml), 1x1ml\nL6000-01G: AB (HRP), 1x15ml\nL6000-01H: Substrate Solution (TMB), 1x15ml\nL6000-01J: Stop Solution (1N H2SO4), 1x15ml\nL6000-01K: Dilution buffer 10X, 2x120ml \nL6000-01L Wash Solution 10X, 1x60ml \n\nStorage and Stability:\nStore components at 4 degrees C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
