Matrix Metalloproteinase 13 (MMP-13, Collagenase 3)

Matrix Metalloproteinase-13 (MMP-13) is one of a family of neutral matrix-degrading proteases. MMP-13 along with interstitial collagenase (MMP-1) and neutrophil collagenase (MMP-8) are the only mammalian enzymes capable of cleaving the triple-helical domain of ?brillar or group I collagens, which include the major interstitial collagens type II and III. MMP-13 shows a substrate speci?city favoring degradation of cartilage type II collagen and is expressed at higher levels by osteoarthritic (OA) chondrocytes than by normal chondrocytes, suggesting a key role for MMP-13 in OA. MMP-13, originally cloned from mammary carcinoma tissue, consists of a single polypeptide chain of 54kD.\n\nThis gene is composed of 10 exons and 9 introns and spans over 12.5 kb. The overall organization of the collagenase-3 gene is similar to that of other MMP genes clustered at chromosome 11q22, including fibroblast collagenase (MMP-1), matrilysin (MMP-7), and macrophage metalloelastase (MMP-12), but is more distantly related to genes coding for stromelysin-3 (MMP-11), gelatinase-A (MMP-2), and gelatinase-B (MMP-9), which map outside of this gene cluster. \n\nHuman collagenase-3 (MMP13) is a recently identified member of the matrix metalloproteinase (MMP) family that is expressed in breast carcinomas and in articular cartilage from arthritic patients. The MMP13 gene has been isolated and characterized. This gene is composed of 10 exons and 9 introns and spans over 12.5 kb. The overall organization of the collagenase-3 gene is similar to that of other MMP genes clustered at chromosome 11q22, including fibroblast collagenase (MMP-1), matrilysin (MMP-7), and macrophage metalloelastase (MMP-12), but is more distantly related to genes coding for stromelysin-3 (MMP-11), gelatinase-A (MMP-2), and gelatinase-B (MMP-9), which map outside of this gene cluster. Ab is the preferred antibody for sensitive detection of RAT MMP-13.\n\nApplications:\nSuitable for use in Immunofluorescence, Immunoprecipitation and Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1ug/ml for 2hrs at RT\nImmunoprecipitation (Native Verified): 2ug/mg protein lysate (Use protein G)\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nConditioned, serum-free medium from TPA-treated rat keratinocyte CCL-4 cells.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
