Matrix Metalloproteinase 9, Catalytic Domain, active, Recombinant, Human (MMP-9, Collagenase 4, Gelatinase B)

EC 3.4.24.35\n\nRecombinant matrix metalloproteinase-9 (MMP-9, gelatinase B, 92kD type IV collagenase) cloned from human cDNA. The enzyme consists of residues Phe107-Pro449, which comprises the catalytic/fibronectin domain of human MMP-9, with a C-terminal purification tag. This represents a naturally-occurring active form of MMP-9 which lacks the C-terminal hemopexin domain. Activity toward its targets, such as gelatin, casein, or peptide substrates, is unaffected.\n\nMatrix metalloproteinases are a family of zinc and calcium dependent endopeptidases with the combined ability to degrade all the components of the extracellular matrix. MMP-9 (gelatinase B) can degrade a broad range of substrates including gelatin, collagen types IV and V, elastin and proteoglycan core protein. It is believed to act synergistically with interstitial collagenase (MMP-1) in the degradation of fibrillar collagens as it degrades their denatured gelatin forms. MMP-9 is produced by keratinocytes, monocytes, macrophages and PMN leukocytes. MMP-9 is present in most cases of inflammatory responses. Structurally, MMP-9 maybe be divided into five distinct domains: a pro-domain which is cleaved upon activation, a gelatin-binding domain consisting of three contiguous fibronectin type II units, a catalytic domain containing the zinc binding site, a proline-rich linker region, and a carboxyl terminal hemopexin-like domain. \nRecombinant protein corresponding to human cDNA aa107-449 active expressed in P. pastoris with a C-terminal purification tag. (NM_004994).\n\nSpecific Activity: \n~60-70U/ug. \n\nUnit Definition: One U=100 pmol/min at 37 degrees C using the colorimetric thiopeptolide Ac-Pro-\nLeu-Gly-S-Leu-Leu-Gly-OEt as substrate. Preincubation of MMP-9 catalytic domain at 1.3nM with the broad-spectrum inhibitor G8237, GM6001 at 30nM for 1 hour inhibits enzymatic activity by 97%.\n\nStorage and Stability:\nAliquot to avoid repeated freezing and thawing and Store at -70 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
