Neuron Specific Nuclear Protein (NeuN)

NeuN antibody reacts with most neuronal cell types throughout the nervous system of mice including cerebellum, cerebral cortex, hippocampus, thalamus, spinal cord and neurons in the peripheral nervous system including dorsal root ganglia, sympathetic chain ganglia and enteric ganglia. The immunohistochemical staining is primarily in the nucleus of the neurons with lighter staining in the cytoplasm. The few cell types which are not reactive include Purkinje, mitral and photoreceptor cells. Developmentally, immunoreactivity is first observed shortly after neurons have become postmitotic, no staining has been observed in proliferative zones. The antibody is an excellent marker for neurons in primary cultures and in retinoic acid-stimulated P19 cells. It is also useful for identifying neurons in transplants.\n\nApplications:\nSuitable for use in Immunohistochemistry, Western Blot and Immunocytochemistry. Other applications have not been tested.\n\nRecommended Dilutions:\nImmunohistochemistry (Paraffin): 1:100. Citrate buffer, pH 6.0 may be used for antigen retrieval in rat cerebellum (optimal staining). Immunoreactivity is seen in nuclear staining in neurons in the granular layer. No signal detected in nucleus of Purkinje cells.\nWestern Blot: Recognizes 2-3 bands in the 46-48kD range and possibly another band at ~66kD. \nImmunocytochemistry: 1:10-1:100. Neurons in culture should be permeabilized with 0.1% Triton X-100. N2173 should be diluted in buffer only, without excess protein blocking agents or detergents.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Controls:\nPositive: Brain Tissue\nNegative: Any non-neuronal tissue (e.g. Fibroblast)\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
