p73 exhibits sequence homology to p53's transcriptional activation, DNA binding, and oligomerization domains. However, adenovirus E1B 55K, simian virus 40 T, and human papillomavirus E6 do not physically interact with p73 indicating that the functions of these two proteins may differ under physiological conditions. p73 is expressed in either a full-length form (a) (MW ~80kD) or a shorter (b) (MW ~70kD) mRNA variant, with exon 13 spliced out and is especially designed for a sensitive detection of both p73a and p73b in Immunoprecipitation and Western Blot assays. Transient overexpression of p73 in cells can induce apoptosis and p21, a cellular p53 target gene primarily responsible for p53-dependent cell cycle arrest.\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation and Immunoelectrophoresis (Supershift). Other applications not tested. \n\nRecommended Dilution: \nWestern Blot: 1-4ug/ml detected p73 splice variants in RIPA lysates from HFF, HEK293, CHO, A431 and Cos-1 cells. Endogenous expression of p73 is low in many lysates but can be induced by ionizing radiation.\nImmunoprecipitation: Immunoprecipitates both p73a and p73b under non-denaturing conditions. The ability to immunoprecipitate p73a is diminished under denaturing conditions.\nOptimal dilutions to be determined by the researcher. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
