Smad4 (DPC4, Small Mothers Against Decapentaplegic Deleted in Pancreatic Carcinoma)

DPC4 (also named SMAD4; small mothers against decapentaplegic deleted in pancreatic carcinoma) belongs to the family of SMAD molecules comprising essential mediators of transforming growth factor beta (TGF-b) signalling. To date, seven members of this family have been identified, each of which plays a specific and separate role in mediating TGF-beta superfamily gene transcription. At least two different members, Smad2 and Smad4/DPC4, have been implicated in human cancer and appear to have tumor-suppressor functions whose inactivation may play an important role in the histogenesis of pancreatic and colorectal malignancy and possibly other human cancers as well.\n\nApplications and Suggested Dilutions: \nImmunofluorescence\nImmunoprecipitation: Use Protein G at 2ug/mg protein lysate.\nWestern Blotting: 1ug/ml for 2hrs at RT\n\nOptimal dilutions to be determined by the researcher.\n\nCellular Localization: \nNuclear\n\nPositive Control: \nLS174T, HT29, Raji, Jurkat cells or pancreatic and colorectal tumors.\n\nHybridoma:\nProduced by the fusion between BALB/c splenocytes and mouse myeloma NS2 cells. \n\nMolecular Weight: 60kD\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
