Src (pp60Src, pp60c-Src)

Src is a member of the Src family of protein tyrosine kinases. This kinase is expressed in different tissues of the body with the highest protein levels detected in neurons and platelets. Src can modulate the signal transduction pathways activated by several growth factors (e.g., PDGF, M-CSF and G-CSF) and integrins. Src also regulates the activity of several ion channels including the N-methyl-D-aspartate (NMDA) receptor. In addition, Src is thought to play a role in physiological/pathophysiological processes in the central nervous system. It has also been reported that mice deficient in Src develop severe osteoporosis. A total of eight membrane-associated protein tyrosine kinases have been identified within the Src gene family. \n\nApplications:\nSuitable for use in Immunoprecipitation and Western Blot analysis. Other applications have not been tested.\n\nRecommended Dilution:\nImmunoprecipitation (kinase): 4ug; Immunoprecipitates Src from 500ug of A431 RIPA lysate. \nWestern Blot: 0.5-2mg/ml. Detects Src in RIPA lysates from non-stimulated A431 carcinoma cells. Previous lots detected Src in RIPA lysates from EGF-stimulated A431 cells, mouse 3T3/A31 cells and Cos-1 cells transfected with pUSE Mouse Src.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control Included:\nA0001-50: A431 Cells, Non-Stimulated in RIPA Buffer, Sample Buffer. Add 2.5ul of 2-mercapoethanol per 100ul of lysate and boil for 5 minutes. Use 20ug per lane for minigels.\n\nStorage and Stability:\n\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
