STAT 4 (Signal Transducer and Activator of Transcription 4)

Signal transducer and activator of transcription 4 (STAT4), also known as gamma interferon activation site-binding protein, is an 86 kD protein that serves as signal transducer and transcriptional activator of the cytokine-induced JAK-STAT pathway (1). This pathway is initiated with the binding of cytokine to its corresponding cell surface membrane receptors (2). This is followed by the association with the cytoplasmic domain of cytokine receptor and its subsequent phosphorylation by JAKs, a family of cytoplasmic tyro-sine kinases. STAT4 has a dual function. It is responsible for both signal transduction and activation of transcription and is expressed in early myeloid differentiation. Defects in the Jak-STAT pathway have already been identi-fied in a number of human diseases such as leukemias, lymphomas and im-munodeficiency syndromes. It can be expected that additional JAK-STAT related diseases will be identified over the next years. To date, specific STAT inhibitory drugs are not known, but a number of specific protein-protein in-teractions in the JAK-STAT pathway are potential targets for pharmaceutical interventions (3).\n\nApplications:\nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot (Colorimetric and ECL): 1ug/ml\nImmunoprecipitation: 10ug/mg \nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nRat Testis Tissue Extract \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
