Thrombospondin (TSP) (BSA & Azide Free)

Thrombospondin is a protein from platelet a-granules. It is made up of three identical subunits bound by interchain disulfides. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells. \n\nApplications:\nSuitable for use in Electron Microscopy, Flow Cytometry, \nImmunofluorescence, Immunoprecipitation, Western Blot and blocks binding of TSP-1 to IAP/CD47. Not suitable for Immunohistochemistry. Other applications not tested. \n\nRecommended Dilution: \nImmunoprecipitation (Native verified): 2ug/mg protein lysate, Use Protein G.\nWestern Blot: 1-2ug/ml for 2hrs at RT, Non-reduced only.\nBlocks Binding of TSP-1 to IAP/CD47: 25-50ug/ml\nOptimal dilutions to be determined by the researcher. \n\nCellular Localization: \nSecretory granules, Golgi complex, endoplasmic reticulum, extracellular matrix\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n

Additional Info

Recognizes Thrombospondin. Shows no crossreactivity with fibronectin, fibrinogen or von Willebrand factor. It blocks TSP-mediated agglutination of fixed RBCs. It shows no effect on TSP-mediated agglutination of fixed, activated platelets. It inhibits both thrombin -and A23187-induced aggregation of washed, live (not fixed) platelets without affecting the secretion of serotonin. Inhibits adhesion of melanoma G361 cells to TSP. Blocks binding of C-terminal domain to Integrin-Associated Protein (IAP)/CD47. Species Crossreactivity: Human, bovine, porcine, equine, canine, sheep and rat.
