Thrombospondin (TSP) (BSA & Azide Free)

Thrombospondin is a protein from platelet a-granules. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells. \n\nApplications:\nSuitable for use in Electron Microscopy, Flow Cytometry. Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested. \n\nRecommended Dilutions: \nImmunoprecipitation (Native verified): 2ug/mg protein lysate, Use Protein G.\nImmunohistochemistry (Formalin/paraffin): 1:25-1:50 for 20 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes.\nWestern Blot: 1-2ug/ml for 2 hours at RT. \nOptimal dilution determined by the researcher. \n\nPositive Control: \nPlatelets in placenta, megakaryocytes in bone marrow \n\nCellular Localization:\nSecretory granules, Golgi complex, endoplasmic reticulum, extracellular matrix\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human Thrombospondin. Shows no crossreactivity with fibronectin, fibrinogen and von Willebrand factor. Shows a mild crossreaction with TSP 2. Inhibits TSP-collagen interaction. Antibody binding to TSP is unaffected by glycosaminoglycans (e.g. hyaluronic acid, chondroitin sulfate and heparin). Its binding is enhanced by EDTA i.e. at low concentration of Ca2+. Species Crossreactivity: bovine, porcine, equine, canine, sheep, mouse and rat.
