TLR9 (Toll Like Receptor 9, CD289) (FITC)

The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules (1) and could constitute an important and unrecognized component of innate immunity in humans. The TRL family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition (2). TLRs characterized so far activate the MyD88/interleukin-1 receptor-associated kinase (IRAK) signaling pathway. Ten human homologs of TLRs (TLR1-10) have been described (3). Gene knockout experiments suggest that TLR9 acts as a receptor for unmethylated CpG dinucleotides in the bacterial DNA (4). Human and mouse TLR9 share an overall amino acid identity of 75.5%. TLR9 is highly expressed in spleen.\n\nApplications: \nSuitable for use in Western Blot, Flow Cytometry and Immunohistochemistry (Frozen and paraffin sections). Other applications not tested. \n\nRecommended Dilution: \nWestern Blot: 1-3ug/ml; In human PBMC and intestine cell lysate a ~120kD band is observed.\nFlow Cytometry: 2-5ug/1x10e6 Ramos cells for intracellular staining.\nImmunohistochemistry: 10-20ug/ml (Frozen sections)\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nHuman PBMC or human intestine cell lysate (WB); Ramos cells (FC).\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
