Troponin I, one of 3 subunits that form the troponin complex of the thin filaments of striated muscle, is the inhibitory component of troponin, the thin filament regulatory complex in striated muscle. Separate genes encode cardiac-specific fast and slow skeletal-specific isoforms of this protein (1). Unlike other contractile proteins, the cardiac isoform of troponin I (TnIc) is expressed only in cardiac muscle and therefore offers a model for cardiac-specific expression. It is also subject to developmental regulation with increased expression occurring at the time of birth (2). TnIc, a sensitive and specific marker of myocardial cell injury, is useful in diagnosing and assessing prognosis in acute coronary syndromes. Studies report that TnIc is elevated in severe heart failure and may predict adverse outcomes (3). Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1,000,000 Immunohistochemistry: 1:250- 1:500 Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
