Tryptophan Hydroxylase, Tyrosine Hydroxylase & Phenylalanine Hydroxylase (TH, TYH, TRH, TYH, PAH, PH8, Tyrosine 3 Hydroxylase, Tyrosine 3 Monooxygenase)

Tyrosine hydroxylase (TYH) is the enzyme which converts tyrosine to dihydroxyphenylalanine (L-dopa), a precursor of the catecholamine neurotransmitters dopamine, noradrenaline and adrenaline. Tryptophan hydroxylase (TRH) is the enzyme that converts 5-hydroxytryptophan to serotonin. \nIn fresh tissue, the antibody binds to TYH and TRH. So it is a marker for TYH-containing catecholaminergic neurons and serotonergic neurons. Tryptophan hydroxylase (TRH) can be used as a marker for serotonin as it converts 5-hydroxytryptophan to serotonin. Serotonin is rapidly metabolized and is unable to be detected by anti-serotonin antibodies in post mortem tissue. In human tissue that has been formalin-fixed, due to a change in the antigenic determinant of tyrosine hydroxylase (TYH), the antibody binds only tryptophan hydroxylase (TRH). Used to specifically identify serotonergic neurons in fixed human tissue. \nPhenylalanine hydroxylase (PAH) can also be detected in hepatic tissue sections. \n\nApplications: \nSuitable for use in the identification of dopaminergic and serotonergic neurons by Immunohistochemistry. Also suitable for Western Blot, Immunoprecipitation and Immunohistochemistry of TYH, PAH and TRH. Other applications not tested.\n\nRecommended Dilution:\nImmunohistochemistry: 1:1000-1:5000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
