The Ku heterodimer comprised of Ku70 and Ku80 subunits is an ATP-dependent DNA helicase II (Lupus Ku autoantigen), involved in nonhomologous end-joining (NHEJ) in mammalian cells. It also plays an important role in V (D) J recombination, apoptosis, telomere fusion, and double-strand break repair, thus contributing to genomic stability. It recruits a 465kD DNA-dependent protein kinase catalytic subunit (DNA-PK) to the DNA double-strand break sites stimulating the DNA-PK kinase activity. Ku70 is the 70kD subunit of the Ku heterodimer that is mainly responsible for the DNA binding activity. It contains two DNA-binding domains, one is located at the N terminus before residue 440, and the other one is located at the C terminus. The DNA binding activity of the C-terminal DNA-binding domain is independent of the heterodimer formation with Ku80, whereas the DNA binding activity of the N-terminal domain of Ku70 depends on heterodimer formation with Ku80. It interacts with Bax and blocks the mitochondrial translocation of Bax. Large amounts of autoantibodies to Ku70 are observed in patients suffering form systemic lupus erythematosus (SLE) and related disorders.\n\nApplications:\nSuitable for use in ELISA, Western Blot, Immunofluorescence and Immunohistochemistry. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
