Additional info: | One of the hallmark features of cells undergoing apoptosis is the formation of membrane blebs in which parts of the plasma membrane protrude, eventually resulting in cells that look like raspberries. As part of the process, membrane phospholipids such as phosphatidylserine and phosphatidylethanolamine redistribute from the inner to outer leaflet of the membrane bilayer where they are exposed on the cell surface. In addition, some nuclear autoantigens translocate to the cell periphery and become clustered in blebs. Cayman's Apoptotic Blebs Assay Kit employs a recombinant protein derived from single-chain variable fragments (scFv) of an antibody which preferentially binds to the autoantigen in membrane blebs of apoptotic cells. This recombinant protein is fused to protein A allowing visualiztion using fluorescein-conjugated rabbit IgG. Since membrane blebbing occurs only in cells committed to cell death, this assay is specific for cells in the execution stage of apoptosis. Propidium iodide is included in the kit for detection of dead cells, through either a necrotic or apoptotic pathway. The kit is easy to use and can be easily adapted to high throughput screening for therapeutic compounds regulating apoptosis. |
