BTF3 (Basic Transcription Factor 3)

The basic transcription factor, BTF3, is a general transcription factor that has been purified from HeLa human cervical carcinoma cell extracts, and is shown to be required for the accurate initiation of transcription from the adenovirus-2 major late promoter (AD2MLP) and other RNA polymerase class B promoters. BTF3 protein exists in two forms, BTF3a, a 206 amino acid long form, and BTF3b, a shorter form lacking the first 44 amino acid residues. BTF3, an apoptosis-associated protein in the human BL60-2 Burkitt lymphoma cell line, is essential for postimplantation development. An insertional mutation in the BTF3 gene leads to an early postimplantation lethality in mice.\n\nApplications:\nSuitable for use in Immunoprecipitation and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nImmunoprecipitation: 7ug/reaction\nWestern Blot: 1-3ug/ml\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Reacts with the human and mouse BTF3 protein. On Western blots, it identifies two bands at ~23kD and ~18kDa, corresponding to BTF3a and BTF3b respectively. A band of unknown origin at ~40kD may be observed. Reactivity has been confirmed with human T47D, human MCF-7, human MDA-MB-468, human HepG2, human U-2 OS, human HL-60, mouse JAWS II cell lysates, and mouse testis or liver homogenetes.
