CD146 (Mucin 18, MUC18 Glycoprotein, Melanoma Associated Antigen A32, S-endo 1 Endothelial Associated Antigen, Melanoma Adhesion Molecule Mel-CM)

CD146 (melanoma-associated antigen A32 or MUC18) is a transmembrane glycoprotein expressed on endothelial cells and is located at the intercellular junction where it plays a role in cell adhesion, and in the cohesion of the endothelial monolayer. CD146 engagement initiates an outside-in signaling pathway involving the protein tyrosine kinases FYN and FAK as well as Paxillin. In addition, it is a signaling molecule involved in the dynamics of actin cytoskeleton rearrangement. CD146 is associated with tumor progression and the development of metastasis in human malignant melanoma. Expression of the antigen is restricted to advanced primary and metastatic melanomas and to cell lines of the neuroectodermal lineage.\n\nApplications:\nSuitable for use in Immunoblot, Immunohistochemistry, Immunoprecipitation and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1-2ug/ml detected MEL-CAM lysates from SK-Mel-37 melanoma cell lysates. SK-Mel-37 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-MEL-CAM (MUC18, CD146) (1ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.\n\nImmunohistochemistry: Detects MEL-CAM in human melanoma samples by PAP immunohistochemistry (1).\n\nImmunoprecipitation: Reported by an independent laboratory to immunoprecipitate MEL-CAM from WM1366 melanoma cell lysates (1).\n\nImmunocytochemistry: Detects MEL-CAM in human keratinocytes (2).\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
