CD59 (MACIF, MIRL, Protectin) (PE)

Human CD59 is a GPI anchored molecule that protects cells from complement-mediated lysis (1). Studies are progressing to determine the regulatory mechanism for CD59 expression (2).\n\nApplications: \nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 1:50 (~80ul/test)\nOptimal dilutions to be determined by the researcher.\n\nFACS Protocol:\n1. Five x 10e5 cultured HPB-MLT cells were washed and preincubated ~5 minutes with 20ul of 250mg/ml human IgG (to block nonspecific binding). \n2. After which they were incubated 45 minutes on ice with 80ul of C2413-21R at a dilution of 1:50. \n3. Cells were washed three times, fixed and analyzed using a BD FACS Calibur. \n4. Cells stained positive with a mean shift of 2.81 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. \n5. Binding was 92% blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml C2413-21N.\n\nStorage and Stability:\nMay be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4 degrees C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
