Chondroitin Sulfate Proteoglycan (NG2, CSPG)

NG2 is a high molecular weight, integral membrane chondroitin sufate proteoglycan. It is found on the surfaces of an unusual class of glial cells within the developing and mature central nervous system that have the properties of oligodendrocyte precursor cells (O-2A progenitor cells). NG2 is also found on the surfaces of chondroblasts, proliferating capillary endothelial cells, some human melanoma cell lines, and on leukemic blasts in childhood acute lymphoblastic leukemia. The NG2 proteoglycan is likely to play a role in regulation of cell motility, axon outgrowth and the cellular responses to platelet derived growth factor.\n\nApplications:\nSuitable for use in Western Blot, Immunocytochemistry, Immunohistochemistry, Immunoprecipitation and ELISA. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:1000. Detects NG2 chondroitin sulfate on 10ug of rat brain lysates.\nImmunocytochemistry: 1:150-1:600\nImmunohistochemistry: 1:200 on frozen embryonic mouse brain tissue using an Alexa Fluor labeled secondary antibody. It is suggested that the tissue used is only lightly fixed (2-4% paraformaldehyde, 5-10min). Avoid overfixing tissue. \nImmunoprecipitation: 2ug/ml \nELISA: 1:1500-1:3000\nOptimal dilution determined by the researcher.\n\nRecommended Positive Control: \nHuman melanoma, glioma and proliferating brain endothelial cells, rat B49 glial cell extracts or embryonic mouse brain tissue.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes rat NG2 Chondroitin Sulfate Proteoglycan between ~270-300kD. Reacts with both the intact Chondroitin Sulfate Proteoglycan and the core protein by Western Blot and ELISA. When oligodendrocyte pecurser cells (O-2A progenitor cells) are stained alive, the stain appears as clusters on the cell surface. Does not stain diferentatied oligodendrocytes well. \nSpecies Crossreactivity: human, mouse and monkey.
