CHOP is a small nuclear protein that is capable of dimerizing with transcription factors C/EBP a and b. Once dimerized, this complex inhibits the normal binding and function of C/EBP to classical binding sites. Inversely, the C/EBP-CHOP dimer gains binding activity to other non-classical C/EBP stress related targets. Under normal cellular conditions this protein is not expressed in detectable levels, but is highly unregulated during times of cellular/ER stress. Examples of CHOP inducing stress include: treatment with tunicamycin, nutrient starvation and reducing agents that interfere with the calcium flux across the ER membrane. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
