Additional info: | The carbohydrate response element-binding protein (ChREBP) is a transcription factor that plays a critical role in the nutrient and hormonal regulation of genes encoding enzymes of glucose metabolism and lipogenesis. Under conditions of low glucose, ChREBP is phosphorylated, which sequesters the transcription factor in the cytoplasm. Upon stimulation with glucose the protein is dephosphorylated at Ser196 and then translocated to the nucleus. The activation of ChREBP has been proposed as a potential mechanism for reducing obesity and improving glucose tolerance. Thus, identification of ChREBP activators is of great interest for drug discovery. The distinct translocation of the protein from the cytoplasm to the nucleus during activation makes it possible to study modulators of ChREBP through sub-cellular localization of the protein using conventional immunocytochemical staining with a specific antibody.
