CIS (SOCS, Suppressor of Cytokines Signaling)

The eight members of the recently identified Suppressor of Cytokines Signalling (SOCS) family are SOCS-1, SOCS-2, SOCS-3, SOCS-4, SOCS-5, SOCS-6, SOCS-7, and CIS. Structurally the SOCS proteins are composed of an N-terminal region of variable length and amino acid composition, a central SH2 domain, and a C-terminal motif called the SOCS box. The SOCS proteins appear to form part of a classical negative feedback loop that regulates cytokine signal transduction. Transcription of each of the SOCS genes occurs rapidly in vitro and in vivo in response to cytokines, and once produced, the various members of the SOCS family appear to inhibit signalling in different ways (1,2).\n\nSOCS-1 and SOCS-6 interact with the insulin receptor (IR) when expressed in human hepatoma cells (HepG2) or in rat hepatoma cells overexpressing human IR. SOCS-1 and SOCS-6 inhibit insulin-dependent activation of ERK1/2 and protein kinase B in vivo and IR-directed phosphorylation of IRS-1 in vitro. These results suggest that SOCS proteins may be inhibitors of IR signalling and could mediate cytokine-induced insulin resistance and contribute to the pathogenesis of type II diabetes (3).\n\nSOCS-6 and SOCS-7 are expressed ubiquitously in murine tissues and SOCS-6 knockout mice are growth retarded (4).\n \nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:1,000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
