FAK is a cytoplasmic non-receptor protein-tyrosine kinase found concentrated in the focal adhesions that form between cells growing in the presence of extracellular matrix constituents and is involved in signaling pathways involved in cell motility, proliferation, and apoptosis. It is activated by tyrosine phosphorylation in response to either integrin clustering induced by cell adhesion or antibody cross-linking, or via G-protein coupled receptor (GPCR) occupancy by ligands such as bombesin or lysophosphatidic acid, or via LDL receptor occupancy. FAK plays a potential role in oncogenic transformation resulting in increased kinase activity. Human FAK gene is localized on chromosomal region 8q24-qter. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Immunohistochemistry (paraffin): 1:50-1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
