FK506 Binding Protein (FKBP59, HSP56, p59)

The 59kD FK506 Binding Protein, FKBP59, was initially characterized as a component of steroid receptor complexes (1). FKBP59 has been identified as a member of the Hsp90/Hsp70 steroid receptor complex, which has been used to differentiate between the active and inactive forms of steroid receptors (2). FKBP59 is alternatively termed FKBP52, HBI and Hsp56 in literature. FKBP59 is an immunophilin of the FK506 binding type whose cellular function is not fully known, but is thought to play a role in the folding and cytoplasmic/nuclear distribution of steroid receptors (3). FKBP59 (Hsp56) exhibits stress protein properties and has been shown to have increased expression in response to heat shock and ischemia (4).\n\nApplications:\nSuitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry, Immunohistochemistry, RIA, ELISA and as a Blocking Antibody. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot (Colorimetric): 2ug/ml. Detects ~59kD band. Additional unidentified 50kD band observed in some cell lysates.\nImmunoprecipitation: 10ug/ml\nImmunohistochemistry: 1:3000\nOptimal dilutions to be determined by researcher.\n\nPositive Controls:\nHeLa Heat Shocked Cell Lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
