Flt3, Cytoplasmic (Fms-like Tyrosine Kinase 3, CD135)

Flt3 is a class III receptor tyrosine kinase that is closely related to c-Kit and c-Fms. It is expressed in primitive hematopoietic cells and its ligand exerts proliferative effects on these cells in vitro in cooperation with other cytokines. Flt3 is also expressed in the placenta, testis, spleen, thymus, bone marrow, brain and in cerebellar Purkinje cells.\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 0.5-2ug/ml detected Flt3 in RIPA lysates from HeLa cells and mouse Bafl-3 cells. Bafl-3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with F4600-08 (1ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. \nImmunoprecipitation: 4ug immunoprecipitated Flt3 from 500ug of mouse Bafl-3 RIPA lysate. \nImmunocytochemistry: 10ug/ml showed positive immunostaining for Flt3 in mouse Bafl-3 cells fixed with methanol and permeabilized with 0.25% Triton X-100 for 5 mins. \nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies:\nWestern Blot:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb\nI1904-46Q: IgG, H&L (HRP) Pab Gt xRb\nImmunocytochemistry:\nI1904-40D: IgG, H&L (FITC) Pab Gt xRb\nI1904-41: IgG, X-Adsorbed (FITC) Pab Gt xRb\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
