Follicle Stimulating Hormone (FSH)

Applications:\nSuitable for use in Immunofluorescence and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilutions:\nImmunofluorescence: 1:20-1:40 in PBS.\nImmunohistochemistry: 1:1000-1:2000 in PBS. Formalin-fixed and paraffin-embedded sections of anterior pituitary glands. Cryostat sections of the same tissue can also be used\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. \n\nImmunofluorescence Staining:\n1. Hydrate the sections with PBS, pH 7.4. \n2. Incubate the section with F5900-16 diluted 1:20-1:40 in PBS at 37 degrees C for 30 minutes. \n3. After washing off the excess antibody with PBS, treat the section with fluorescein labeled GtxRb IgG for 30 minutes at 37 degrees C. Wash the tissue with PBS and mount under glycerine-saline.\n\nImmunoperoxidase Staining (IHC):\n1. Hydrate the sections with PBS, pH 7.4. \n2. Incubate the section with F5900-16 diluted 1:1000-1:2000 in PBS at 37 degrees C for 30 minutes. \n3. Wash for 15 minutes with PBS at RT and treat sequentially with GtxRb IgG and PAP complex at 37 degrees C. Wash excess reagent from the section as described above. \n4. Treat the section with a mixture of 3,3'-diaminobenzidine HCl (0.02%) in hydrogen peroxide (0.004%). For preservation, the sections may be dehydrated, cleared and mounted.
