Heparin Cofactor II (HCII) BioAssay(TM) ELISA Kit (Antibodies only)

Heparin Cofactor II (HCII) is a plasma glycoprotein that is a member of the SERPIN family of proteinase inhibitors. HCII is a relatively specific inhibitor of thrombin and its activity is increased approximately 1000 fold in the presence of heparin. Its activity is also increased in the presence of dermatan sulfate. HCII is a single chain molecule with a mass of 65.6kD. Inhibition of thrombin results in a covalent complex of 102.6kD. HCII in plasma and other fluids can be measured by capturing onto a microtitre plate coated with an affinity purified antibody to HCII. After washing the plate to remove unbound material, a peroxidase conjugated second antibody is added to the plate which will bind to the captured HCII.\n\nAfter washing of the plate, the peroxidase activity is expressed by incubation with ortho-phenylene diamine (OPD) and after a fixed time the reaction is quenched with the addition of H2SO4. The color generated is proportional to the concentration of HCII present in the sample.\n\nMaterials Supplied:\nAntibodies for 4x96 well plates\n\nKit Components:\nH11892-60A: Capture Antibody \n1x400ul Affinity Purified Supplied as a liquid. For coating plates.\n\nH11892-60B: Detecting Antibody (HRP) 4x10ml Affinity Purified Supplied as a liquid. For detection of captured HCII.\n\nStorage and Stability:\nMay be stored at 4 degrees C. For long-term storage, aliquot and store at 4 degrees C. Do not freeze. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
