Heregulin alpha-1, Recombinant, Human (HRGa1)

Heregulin (HRG) is a glycoprotein of 44kD which binds to c-erbB-3 and c-erbB-4 receptors with low and high affinities, respectively. Heregulins constitute a family of mosaic proteins comprising of at least 12 members. They contain certain recognizable domains which include: an amino terminal (N-ter) region; an immunoglobulinlike motif (Ig); a glycosylated spacer sequence (glyco); an EGF-like motif (EGF), which is subdivided into a and b isoforms; a juxtamembrane region subdivided into isoforms 1-3; a hydrophobic transmembrane domain (TM); a common cytoplasmic tail (Cyto); and a variable-length cytoplasmic tail (a-c). Using this nomenclature, several isoforms of HRG are designated as HRG-a1, HRG-a2, HRG-a3, HRG-b1, HRG-b2, HRG-b3, HRG-a2b, etc. Note: Though the neuronal counterparts of heregulin, (ARIA and GGF), are similar they do not have the spacer sequence and have a variable N-terminal region. GGF II contains a putative kringle motif and a hydrophobic signal peptide in addition.\n\nComments: Extracellular domain of Heregulin a1 was expressed in E. coli. In SDS-PAGE, the purified recombinant Heregulin a1 protein appears as a single band at 30kD. Heregulin a1 protein modulates the proliferation and differentiation of cultured mammary cells and stimulates the tyrosine kinase activity of ErbBs.\n\nApplications:\nSuitable for use in Western Blot,\nStimulation of Tyrosine Kinase Activity of Erbs, Cell Proliferation and Differentiation. Optimal dilutions to eb dtermined by the researche\n\nPositive Control: T47-D breast cancer cells\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
