Histone H3, acetylated (Lys14)

Histones in the nucleosome core (H2A, H2B, H3 and H4) possess a positively charged unstructured amino termini domain, which are targets for post-translational modifications. Histone H3 modifications include the acetylation and phosphorylation of specific residues in the amino terminal domain. The acetylation of the e-amino group of lysine residues is regulated by an emerging family of enzymes called histone acetyl transferases (HATs) and deacetylases that exhibit site and histone specificity. Histone acetylation is believed to weaken histone:DNA interaction thereby facilitating access of the transcriptional machinery. The phosphorylation of core histone H3 occurs at serine residue10, following mitogen stimulation and during all phases of mitosis. Acetylation of histone H3 at lysine residue 14 is the preferred site for the yeast transcriptional adapter GcN5p as compared to the lysine at residue 9.\n\nMolecular Weights of Histones:\nLysine Rich (H1, f1): ~ 21,500 \nSlightly Lysine Rich (H2a, f2a2): 14,004 \nSlightly Lysine Rich (H2b, f2b): 13,774 \nArginine Rich (H3, f3): 15,324 \nArginine Rich (H4, f2a1): 11,282\n\nApplications:\nImmunoblot Analysis: 0.1ug/ml detects acetylated histone H3 in acid extracted proteins from HeLa cells. Sodium butyrate, an inhibitor of histone deacetylases, was used to enhance detection of acetylated histone H3. Acid-extracted proteins from normal HeLa cells and HeLa cells treated with sodium butyrate were resolved by electrophoresis, transferred to nitrocellulose and probed with Anti-Acetyl-Histone H3 (Lys 14), (0.1mg/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. \n\nImmunoprecipitation: 1ug immunoprecipitates acetylated histone H3 from 500ug of an acid precipitate of sodium butyrate treated HeLa cells. \n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
