Histone H3, phosphorylated (Ser10) (Agarose)

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the phosphorylation of serine or threonine. Phosphorylation at Ser10 (pSer10), which is linked to gene activation, prevents methylation at Lys9 but facilitates acetylation of H3 and H4. pSer10 is crucial for chromosome condensation and cell-cycle progression during mitosis and meiosis. In addition, Ser10 phosphorylation is also an essential regulatory mechanism for neoplastic cell transformation. Phosphorylation at Ser10 by Aurora-B mediates the dissociation of HP1 proteins (CBX1, CBX3 and CBX5) from heterochromatin.\n\nApplications:\nSuitable for use in Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nImmunoprecipitation: 20ul of a 50% slurry.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
