Histone H4, acetylated

Applications:\nWestern Blotting: 1:2000 dilution detects acetylated histone H4 in Tetrahymena macronuclei and acid extracted proteins from HeLa cells treated with 5mM sodium butyrate. Sodium butyrate, an inhibitor of deacetylases, was used to enhance detection of acetylated histone H4.\nChromatin Immunoprecipitation: 5-10ul immunoprecipitates transcriptionally active\nchromatin containing acetylated histone H4 from 2X 106 serum stimulated HeLa cells.\nImmunocytochemistry: Not recommended. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes acetylated histone H4 of approximately 10kD. Cross-reacts with acetylated\nhistone H2B from Tetrahymena and weakly crossreacts with acetylated histone H2B from HeLa cells. May cross-react with other acetylated proteins. Crossreactivity: Human and Tetrahymena. Other species not tested, but expected to crossreact since Histone H4 is well conserved.
