Histone H4, trimethyl (Lys20)

Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H4 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H4 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.\n\nApplications: \nSuitable for use in Western Blot, Peptide Inhibition, Chromatin Immunoprecipitation and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot:1:500-1:2000 detected methylated histone H4 in acid extracted proteins from HeLa cells, but not demethylated recombinant Histone H4.\nPeptide Inhibition: Specificity confirmed by the ability of 1mM of the immunizing peptide to completely abolish detection of Histone H4 in Western Blot analysis of HeLa acid extracts. Slight signal reduction was detected with preincubation of this lot with 1mM dimethyl-lysine 20.\nChromatin Immunoprecipitation: Has been reported to immunoprecipitate chromatin containing trimethylhistone H4 (Lys20).\nImmunocytochemistry: Has been reported to detect trimethylated histone H4 in mouse embryonic fibroblasts.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
