Interleukin 1 beta (IL-1b)

Interleukin-1B (IL-1beta) is a potent immuno-modulator which mediates a wide range of immune and inflammatory responses including the activation of B and T cells. Human IL-1beta is a 17kD protein containing 153 amino acid residues.\n\nApplications: \nSuitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization of Bioactivity. Other applications not tested.\n\nRecommended Dilution:\nDirect ELISA: 0.5-1ug/ml. Detection limit for rhIL-1b is ~0.15ng/well.\nWestern Blot: 0.1-0.2ug/ml. Detection limit for rhIL-1b is ~0.5ng/lane under both non-reducing and reducing conditions.\nImmunohistochemistry (Frozen): 5-15ug/ml. Detects human IL-1b in cultured cells or tissue sections.\nOptimal dilutions to be determined by the researcher.\n\nNeutralization of Human IL-1b Bioactivity:\nThe exact concentration of antibody required to neutralize rhIL-1b activity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. To provide a guideline, the neutralization dose for this antibody can be determined under a specific set of conditions. The Neutralization Dose 50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when that cytokine is present at a concentration just high enough to elicit a maximum response. The ND50 for this lot of I7663-11C was determined to be ~0.004-0.008ug/ml in the presence of 50pg/ml of recombinant human IL-1b using a D10.G4.1 cell proliferation assay.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Will not neutralize the biological activity of recombinant mouse IL-1b, recombinant human IL-1a, or recombinant mouse IL-1a. Based on direct ELISA results, this antibody shows less than 10% crossreactivity with recombinant mouse IL-1b, recombinant porcine IL-1b, recombinant rat IL-1b, recombinant crIL-1b and recombinant rfeIl-1b.
