Interleukin-19 (IL-19) is a novel cytokine that was initially identified during a sequence data base search aimed at finding potential Interleukin-10 homologs. Interleukin-19 shares 21% amino acid identity with IL-10. The exon/intron structure of IL-19 is similar to that of the human IL-10 gene, comprising five exons and four introns within the coding region of the IL-19 cDNA. IL-19 shares a receptor complex with IL-20, indicating that the biological activities of these two cytokines overlap and that both may play an important role in regulating development and proper functioning of the skin. Unlike IL-10, which forms an intercalated dimer, the molecule of IL-19 is a monomer made of seven amphipathic helices, A-G, creating a unique helical bundle.\n\nDescription: \nRecombinant Human IL-19 produced in E.Coli is a single, non-glycosylated polypeptide chain containing 155 amino acids and having a molecular mass of 17913 Dalton. Recombinant Human Interleukin-19 is purified by proprietary chromatographic techniques.\n\nAmino Acid Sequence: \nThe sequence of the first five N-terminal amino acids was determined and was found to be Met-Leu-Arg-Arg-Cys.\n\nDimers and Aggregates: \nLess than 1% as determined by silver-stained SDS-PAGE gel analysis.\n\nBiological Activity: \nRecombinant Human Interleukin-19 is fully biologically active when compared to standard. The activity is determined by it's ability to activate STAT following receptor-ligand interaction.\n\nEndotoxin: \n(same/less than) 0.1ng/ug (IEU/ug) of human IL-19.\n\nProtein Content: \nProtein quantitation was carried out by two independent methods:\n1. UV spectroscopy at 280nm. \n2. Analysis by RP-HPLC, using a standard solution of IL-19 as a Reference Standard.\n\nSolubility: \nIt is recommended to reconstitute the lyophilized Interleukin-19 in sterile 18MOmega-cm H2O not less than 100microg/ml, which can then be further diluted to other aqueous solutions.\n\nStability: \nLyophilized Interleukin-19 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution Recombinant Human IL-19 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
