Interleukin 21, Recombinant, Human (IL-21)

Interleukin-21 (IL-21), expressed by activated CD4(+) T cells, is the newest member of the common gamma-chain family of cytokines, which includes IL-2, IL-4, IL-7, IL-9, IL-13, and IL-15. Its private receptor, IL-21R, is expressed on T, B, and NK cells. Within the B cell lineage, IL-21 regulates IgG1 production and cooperates with IL-4 for the production of multiple Ab classes in vivo. IL-21R has been shown to activate the Janus kinase/signal transducers and activators of transcription signaling pathway upon ligand binding. Initial studies have demonstrated that IL-21 has pleiotropic effects on the proliferation, differentiation, and effector functions of B, T, natural killer, and dendritic cells.\n\nDescription: \nRecombinant Human IL-21 produced in E.Coli is a single, non-glycosilated polypeptide chain containing 133 amino acids and having a total molecular mass of 15,463 Dalton. The Recombinant Human IL-21 is purified by proprietary chromatographic techniques.\n\nAmino Acid Sequence: \nThe sequence of the first five N-terminal amino acids was determined and was found to be Gln-Gly-Gln-Asp-Arg.\n\nBiological Activity: \nThe ED50=1-10ng/ml as determined by the dose-dependent proliferation of human peripheral blood mononuclear cells is < 5ng/ml.\n\nProtein Content: \nProtein quantitation was carried out by two independent methods:\n1. UV spectroscopy at 280 nm using the absorbency value of 0.54 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).\n2. Analysis by RP-HPLC, using a standard solution of IL-21 as a Reference Standard.\n\nStorage and Stability: \nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O, 0.1% HSA or BSA, ,aliquot and store at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.\n
