JAK3, a member of the Janus associated family of protein tyrosine kinases plays a crucial role in hematopoietic cytokine signaling. It is associated with the common gamma chain of cytokine receptors and mediates the downstream signaling of cytokines, interleukin IL-2, IL-4, IL-7, IL-9, and IL-15. It is activated upon cytokine binding to the specific cellular receptors, and phosphorylates tyrosine residues of receptor chains which generate docking sites for Src homology 2 (SH2)-containing signal transducers and activators of transcription (STAT) proteins. STAT are in turn phosphorylated before they dimerize and translocate to the nucleus where they bind to promoter sequences of cytokine-inducible genes mediating their transcription. In humans the protein is primarily expressed in hematopoetic cells, natural killer cells and stimulated or transformed T cells. The open reading frame of the JAK3 gene comprises 3372bp, that are organized in 23 exons and translate into a 1124aa protein of approximately 125kD. The protein consists of 7 Janus homology (JH) domains based on sequence homology with other JAKs, which includes an amino-terminal FERM domain (Band 4.1, Ezrin, Radixin, Moesin homology domain), an src homology 2 (SH2)
