LRRK2 (Leucine-rich Repeat Kinase 2, RIP7, PARK8, Dardarin)

This gene is a member of the leucine-rich repeat kinase family and encodes a protein with an ankryin repeat region, a leucine-rich repeat (LRR) domain, a kinase domain, a DFG-like motif, a RAS domain, a GTPase domain, a MLK-like domain, and a WD40 domain. The protein is present largely in the cytoplasm but also associates with the mitochondrial outer membrane. Mutations in this gene have been associated with Parkinson disease-8.\n\nApplications:\nSuitable for use in ELISA, Immunohistochemistry (Paraffin), Immunoflourescence and Western Blot. Other applications not tested. \n\nRecommended Dilution: \nImmunohistochemistry: 1:100 \nImmunofluorescence: 1:1000 (Mouse CAD cells transfected with human wild-type LRRK-2, \nimmunoprecipitation: 7ul/15 cm plate of SH-SY5Y cells grown to 80% conflue, \nWestern Blot: 1:1(the theoretical molecular weight of LRRK2 is ~286kD. Other bands have been seen with some lysates, but these bands have been blocked by the control peptide, L4502-21E, suggesting that these bands are degredation products)\nOptimal dilution determined by the researcher. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n

Recognizes human LRRK2. Species Croossreactivity: Bovine. Negative in mouse. There is 87% homology of the immunogen with the rat LRRK2 proteins. Homology: 100% homology with human protein, 94% with mouse and rat protein.
