Lysine, Acetylated (Acetyl-Lysine)

In the nucleus, DNA is tightly packed into nucleosomes generating an environment which is highly repressive towards DNA processes such as transcription. Acetylation of lysine residues within proteins has emerged as an important mechanism used by cells to overcome this repression. The acetylation of non-histone proteins such as transcription factors, as well as histones appears to be involved in this process. Acetylation may result in structural transitions as well as specific signaling within discrete chromatin domains. The role of acetylation in intracellular signaling has been inferred from the binding of acetylated peptides by the conserved bromodomain. Furthermore, recent findings suggest that bromodomain/acetylated-lysine recognition can serve as a regulatory mechanism in protein-protein interactions in numerous cellular processes such as chromatin remodeling and transcriptional activation.\n\nApplications: \nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:500 on 10ug of sodium butyrated treated HeLa lysates. Proteins were resolved by electrophoresis, transfered to PVDF and probed with L9058-02A. Proteins were visualized using IgG (HRP) goat anti-mouse secondary antibody and chemiluminescence detection system.\nImmunoprecipitation: 5ug immunoprecipitates in vitro acetylated PCAF added to a 3T3 RIPA cell lysate. The immunoprecipitated PCAF was detected by subsequent Western Blot analysis using 1ug/ml G8135-05.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nTSA-treated HeLa and NIH/3T3 cells, osteosarcoma tissue\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
