Applications: \nSuitable for use in Immunoprecipitation and Affinity Purification. Other applications have not been tested.\n\nRecommended Dilution:\nImmunoprecipitation: 2.5-5ug (5-10ul gel slurry) Immunoprecipitate MAP Kinase from 500ug of a RIPA lysate of 3T3/A31cells. Usually after boiling in SDS-PAGE sample buffer only an extremely small amount of the non-covalently bound IgG light chain (~ 28kD) is observed. This precipitated enzyme can be used to phosphorylate myelin basic protein (MBP). The phosphorylated substrate is then analyzed by western immunoblot using an antibody specific for phosphorylated MBP \nAffinity Purification: Antibody-agarose can be re-used a minimum of three times after elution of MAP Kinase R2 by either low pH, high pH, or high salt. MAP Kinase R2 may be released from the antibody-agarose by the following agents: \nOptimal dilutions to be determined by the researcher.\n\nNote: It is recommended to wash the agarose beads with appropriate buffer prior to use to remove sodium azide. \n\nElution Profile: \nElution Buffer Relative % Antigen Released\nSDS-PAGE sample buffer 100\n0.1M Glycine, pH 2.5 50 \n0.1M TEA, pH 11.5 40 \n3.5M MgCl2, pH 7.2 70 \n\nStorage and Stability:\nMay be stored at 4 degrees C.
