An important convergence point involved in the signal transduction pathways of many different growth factors, hormones and cytokines is a family of 41-44kD serine/threonine kinases collectively called either MAPKs (for mitogen activated protein kinases) or Erks (for extracellular regulated kinases). MAPK is activated by sequential phosphorylation on both tyrosine and threonine residues by either the dual tyrosine/threonine kinase Mek or by Mek in conjunction with another unknown kinase. During intracellular signaling, Mek is phosphorylated and activated by the serine/threonine kinase Raf. Activation of MAPK is directly regulated by a specific MAPK phosphatase and indirectly regulated by protein kinase A activation which results in inhibition of Raf activity in mammalian cells. \n\nApplications:\nSuitable for use in Immunoblot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nImmunoblot: 0.5-2ug/ml. Detected pMAPK in RIPA lysates of NGF-stimulated PC-12 cells. Serum-stimulated 3T3/A31 and non-stimulated 3T3/A31 cell lysates were resolved by electrophoresis, transferred to nitrocellulose and probed with M2363-05 (2ug/ml). Proteins were visualized using rabbit anti-sheep secondary antibody conjugated to HRP and a chemiluminescence detection system was used. \nImmunoprecipitation: 4ug immunoprecipitates pMAPK from a 500ug of EGF-stimulated A431 RIPA lysate.\nOptimal dilutions to be determined by reseacher.\n\nRecommended Secondary Antibodies:\nRabbit anti-sheep IgG labeled with HRP\nI1904-59C: IgG, H&L (HRP) Pab Rb xSh\nI1904-59H: IgG, H&L, X-Adsorbed (HRP) Pab Rb xSh\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
