Both p44 and p42 MAP kinases (Erk1 and Erk2) function in a protein kinase cascade that plays a critical role in the regulation of cell growth and differentiation (1-4). MAP kinases are activated by a wide variety of extracellular signals including growth and neurotrophic factors, cytokines, hormones and neurotransmitters. Activation of MAP kinases occurs through phosphorylation of threonine and tyrosine (202 and 204 of human MAP kinase [Erk1] or 183 and 185 of rat Erk2) at the sequence T*EY* by a single upstream MAP kinase kinase (MEK) (5,6). Both kinases are known to weakly autophosphorylate on tyrosine (5).\n\nApplications: \nSuitable for use in Flow Cytometry, ELISA, Western Blot, Immunoprecipitation, Immunofluorescence and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:2000, incubate membrane with diluted antibody in 5% nonfat dry milk, 1X TBS, 0.1% Tween-20 at 4 degrees C with gentle shaking, overnight.\nImmunoprecipitation: 1:500\nImmunofluorescence: 1:200\nImmunocytochemistry: 1:200\nFlow Cytometry: 1:1000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes endogenous levels of human p44 and p42 MAP kinase (Erk1 and Erk2) when dually phosphorylated at Thr-202 and Tyr-204 of Erk1 (Thr-185 and Tyr-187 of Erk2) and singly phosphorylated at Try-204. Does not crossreact with the corresponding phosphorylated residues of either SAPK/JNK or p38 MAP kinase. This antibody may cross-react with an unknown cytoskeletal protein in some cell lines as visualized by immunofluorescence.Species Crossreactivity: Hamster, mouse, rat and zebrafish.
