MSK2 (Mitogen and Stress activated protein Kinase1)

Members of the RSK (the p90 ribosomal S6 kinase) family of serine/threonine kinases are substrates of ERK (the Ras/extracellular signal-regulated kinase). ERK pathway regulates a variety of cellular processes that include cell proliferation, survival, growth and motility. The RSK family consists of four isoforms(RSK1 to -4) and two structurally related cousins, called RSK-like protein kinase (RLPK/MSK1) and RSKB(MSK2). MSK2 is activated by p38- and ERK-mitogen-activated protein kinase pathways. MSK2 C-terminal extension contains elements that control activation threshold, subcellular location, and p38 docking. Similar to other RSKs6, MSK2 is located in the nucleus. MSK2 maps to the BBS1 locu, which is associated with Bardet-Biedl syndrome, with manifestations similar to Coffin-Lowry syndrome, including mental retardation, and thus may be a candidate BBS gene. MSK2 is persistently activated through tumor necrosis factor and selectively escapes more common down regulatory cell mechanisms that cause rapid deactivation of upstream p38 and ERK and other RSKs5. MSK1 and MSK2 are rate limiting for the LPS-induced activation of CEB/ATF1, and the transcription of the COX-2 and IL-1b genes, suggesting that they play a role in these processes and are potential targets for the development of novel anti-inflammatory drugs. Studies using knockout mice show that MSK1 and MSK2 are requited for the mitogen- and stress-induced phosphorylation of CREB and ATF1 in fibroblasts. MSK1 and MSK2 are the major kinases for histone H3 and HMG-14 in response to mitogenic and stress stimuli in fibroblasts. The calculated molecular weight of MSK2 is ~86kDa.\n\nApplications:\nSuitable for use in Immunoprecipitation and Western Blot. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
