Myostatin (GDF8, MSTN) is a member of the transforming growth factor beta superfamily that is essential for proper regulation of skeletal muscle mass. Myostatin is a secreted protein that negatively regulates skeletal muscle mass determining both muscle fiber number and size. The myostatin pathway is conserved and regulates muscle mass in a number of animal species ranging from fish to humans. Inhibition of myostatin using a variety of therapeutic approaches can increase muscle mass in a number of animal models of human disease, including muscular dystrophy. \n\nRecombinant Human Myostatin, also called GDF-8, produced in E. coli, is a homodimer, non-glycosylated polypeptide chain containing 2x109 amino acids and having a total molecular mass of 24.814kD.\n\nAA Sequence:\nThe sequence of the first five N-terminal amino acids was determined and was found to be Asp-Phe-Gly-Leu-Asp. \n\nBiological Activity:\nRecombinant Human Myostatin is fully biologically active when compared to standard. The ED50 as determined by the inhibition of the proliferation of MPC-11 cells is ~ 20ng/ml. \n\nProtein Content:\nProtein quantitation was carried out by two independent methods: \n1. UV spectroscopy at 280 nm using the absorbency value of 1.55 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). \n2. Analysis by RP-HPLC, using a standard solution of Myostatin as a Reference Standard. \n\nApplications: \nSuitable for use in ELISA, Western Blot and Inhibition Assays. Other applications not tested.\n\nRecommended Dilutions:\nOptimal dilutions to be determined by researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Reconstitute with sterile 20mM acetic acid, 0.1% HSA or BSA. Aliquot and store at -20 degrees C. Reconstituted product is stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from a concentrated solution containing no additives. Reconstitute with sterile 20mM acetic acid to a concentation of (same/more than)100ug/ml, which can then be further diluted to other aqueous solutions with 0.1% BSA or HSA. Lyophilized myostatin is poorly soluble at neutral pH.
