NEDD4 (E3 Ubiquitin-protein Ligase NEDD4, Cell Proliferation-inducing Gene 53 Protein, Neural Precursor Cell Expressed Developmentally Down-regulated Protein 4, NEDD-4, KIAA0093, NEDD4-1, PIG53)

NEDD4 (Neural precursor cell-expressed developmentally down-regulated gene 4-1; also RPF1) is a 115-120kD member of the HECT E3 ubiquitin ligase family of proteins. The NEDD4 gene is widely expressed, and serves as a complement to a related gene product termed NEDD4-2/NEDD4L. Both molecules ubiquitinate multiple proteins. NEDD4-1 acts preferentially on PTEN, tumor suppressors and endocytic proteins, while NEDD4-2 acts preferentially on membrane transporters. Human NEDD4-1 is 900aa in length and contains one PKC region (aa20-124), four WW domains (aa196-505) and one E3 HECT domain (aa543-897). Caspase cleavage after Asp197 generates a 95kD C-terminal, and smaller but variable sized N-terminal fragment. There are multiple splice forms. Two show a 516aa substitution for aa1-169 and 1-97, respectively, while a third shows the latter substitution coupled to a deletion of aa170-185. Over aa211-349, human NEDD4-1 shares 83% and 72aa identity to mouse and rat NEDD4-1 and less than 25aa identity with human NEDD4-2. \n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 25ug/ml \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
