PCNA, Recombinant, Human (Proliferating Cell Nuclear Antigen)

Proliferating cell nuclear antigen (PCNA), a protein of 30kD molecular weight also known as cyclin, forms a trimer ring around a DNA double-helix. It binds to a variety of other nuclear proteins and thereby organises biochemical processes at the DNA replication fork. PCNA thereby assumes important functions in DNA synthesis, repair and processing, the regulation of cell cycle and even apoptosis. Anti-PCNA antibodies occur in systemic lupus, where they represent a rare specificity (< 10 % of SLE patients). However, their presence alone or in combination with other autoantibodies should alert the clinician to the presence of defined or evolving SLE.\n\nPCNA Human Recombinant produced in SF9 is a glycosylated, polypeptide chain having a molecular mass of 29.7kD. PCNA is expressed with a -6xHis tag and purified by proprietary chromatographic techniques.\n\nImmunological Functions:\n-Binds IgG-type human auto-antibodies.\n-Standard ELISA test (checkerboard analysis of positive/negative sera panels, immuno-dot analysis with validated SLE patient sera.\n\nApplications:\nSuitable for use in ELISA and Western Blot with monoclonal anti-hexa-His-tag antibody and Scleroderma patient sera. Suitaable for biotinylation and iodination. Other applications not tested. \n\nRecommended Dilutions: \nELISA: 0.5-1.1ug/ml\nOptimal dilution determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
