PHAP I, CT (Putative HLA DR-associated Protein)

Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Cytochrome c, after released from mitochondria, binds to Apaf-1, which forms an apoptosome that in turn binds to and activates procaspase-9. Activated caspase-9 cleaves and activates the effector caspases (caspase-3, -6 and -7), which are responsible for the proteolytic cleavage of many key proteins in apoptosis. The tumor suppressor putative HLA-DR-associated proteins (PHAPs) were recently identified as important regulators of mitochondrion apoptosis (1). PHAP appears to facilitate apoptosome-medicated caspase-9 activation and to stimulate the mitochondrial apoptotic pathway. PHAP was also shown to oppose both Ras and Myc-mediated cell transformation.\n\nApplications: \nSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 0.5-2ug/ml; PHAP I expression in human Raji cells. A band at ~32kD can be detected.\nImmunohistochemistry: 2ug/ml; Stains mouse small intestine tissue.\nOptimal dilutions to be determined by the researcher.\n\nBlocking Peptide: \nP3368-02A: PHAP I, CT, Control Peptide (Putative HLA DR-associated Protein)\n\nPositive Control: \nRaji cell lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
