PHLPP2, a protein phosphatase and negative regulator, which mediates the inactivation of Akt and protein kinase C (PKC) activity, through the direct dephosphorylation of key regulatory sites: AKT1 (Ser473), PRKCA (Ser657) and PRKCB isoform beta-II (Ser660). PHLPP2, and its alternative isoform PHLPP1, have been implicated as important suppressors of tumour cell growth, being linked with breast, ovarian and colon cancer.
Applications:
Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilution:
ELISA: 1:32,000
Western Blot: 0.1-0.3ug/ml, observed in human brain (amygdala, hippocampus, substantia nigra) lysates on ~140kD bands
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
