Phosphoinositide 3 kinase, p110 gamma (PI3K)

Phosphoinositide 3-kinases are a family of enzymes that phosphorylate the D-3 position of phosphoinositides. The products of PI3K-catalyzed reactions act as second messengers for a variety of signal transduction pathways, including those essential to cell proliferation, adhesion, survival, cytoskeletal rearrangement and vesicle trafficking. The mammalian PI3Ks can be divided into three classes based on their structure and substrate specificity. The class I PI3Ks are receptor-regulated heterodimeric enzymes consisting of a 110kD catalytic subunit and an adaptor subunit. Although they can phosphorylate PtdIns, PtdInsP and PtdInsP2 in vitro, their preferred substrate in vivo is PtdInsP2. The class IA PI3Ks (consisting of p110a, p110b and p110d isozymes) associate with an 85kD adaptor subunit that is essential for interaction of these PI3Ks with receptor tyrosine kinases. The class IB PI3Ks (consisting of p110g) are activated by heterotrimeric G protein subunits and associate with a p101 adaptor subunit that is important for full responsiveness to Gbg heterodimers. Both class IA and IB PI3Ks are also activated by Ras. Class II PI3Ks, distinguished by a C-terminal C2 domain, are larger enzymes of ~170kD that preferentially use PtdIns and PtdInsP as substrates. Class III enzymes, such as yeast Vps34p, were originally isolated from mutant yeast strains that had defective vacuolar protein sorting. These enzymes, which only phosphorylate PtdIns, have now been identified in other species.\n\nApplications: \nSuitable for use in Immunoprecipitation and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
