Phosphoserine

Cellular responses to a variety of extracellular signals occur through phosphorylation or dephosphorylation of intracellular proteins. Abnormal protein phosphorylation may be involved in the progression of numerous diseases, including many forms of cancer, immune system dysfunction and cardiovascular disease. Such changes in protein phosphorylation may be detected by immunocytochemical mapping of cells labeled with antiphosphoserine or antiphosphothreonine antibodies and can be correlated with cellular, electrophysiological or behavioral state changes.\n\nApplications:\nSuitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and ELISA. Other applications have not been tested. \n\nRecommended Dilutions:\nWestern Blot:1:500 detects serine phosphorylated proteins on 10ug of NGF PC12 lysates. NGF treated PC12 lysates were resolved by electrophoresis, transferred to PVDF membrane and probed wiht P4076. Proteins were visualized using a donkey anti-rabbbit secondary antibody (HRP) and a chemiluminescence detection system.\nImmunoprecipitation (Tissue extracts): 10-20ug/mg protein. Note that this will immunoprecipitate all phosphoserine proteins in the extracts. If using purified protein exacts containing only the protein of interest, antibody amounts should be decreased to 5ug/500ul of extracts. \nELISA (Kinase assay): 1:250-1:500\nImmunohistochemistry: 1:50\nOptimal dilutions to be determined by the researcher.\n\nPositive Controls:\nNIH 3T3 cells (+/- TPA), K562 cells and EGF-stimulated A431 cells.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes phosphoserine, peptidyl-phosphoserine and serine-phosphorylated proteins. Does not crossreact with ATP, phosphotyrosine, peptidyl phosphothreonine and serine. Slight crossreactivity with free phosphothreonine. Readily reacts to known phosphoproteins such as phosvitin and alpha casein. \nSpecies Crossreactivity: All species.
